Alpha Helix Data page 2 .4. EQUIPMENT-SUPPLIES INVENTORY ON SHIP Forhan is doing OK m n assembling items from the equipment print-out passed around in Kalamazoo. Keep in mind, however, that if it is critical you should bring it yourself!!! This would apply particularly to small, easily overlooked, items or special chemicals. Specific problems or comments on equipment: a. Specimen bottles: bring your own, inventory is short on these. b. Hot plates: there are 2 on the ship. Someone requested 5 more (at $90 each) ! Bring your own. c. Styrofoam boxes -buy in port or load in Florida if needed. -d. SCUBA tanks: bring your own if you can with back pack or ship it to Florida. We can't afford to buy them and there are only 6 on board. e. SCUBA weights; unless you hear otherwise we will supply these, but bring your own belt. f . Freeze-dryer: We are shipping small unit loaned to St. Croix by Pat Parker to the ship. g . Dry ice: We will have syphon CO2 cylinders on board, but make sure you have back-ups on ice makers or another source lined up. 5 . SHIPPING OR HAND-CARRYING SPECIMENS, PLANTS AND SEDIMENT SAMPLES FROM BELIZE OR COLON. Each investigator is responsible for getting the appropriate permits for his hand-carried or freighted collections. 6. It will be possible for some participants in Leg I to ride the ship from Florida to Belize. The sailing date from Florida is about Oct. 7; time en route about 5 days. If you are interested let me know as soon as possible. Forhan does not want any 'riders' to show up at the ship a.) unannounced and b , ) before sailing date. 7. SCRIPPS SCUBA REQUIREMENTS Pull every string that you can to get out of it, but S.I.O. does require that scuba exam along with the medical forms and diving application. They have a diving control board that likes to review this stuff. Tom Forhan assures me that so far there is no one who will have problems with diving from the ship, but keep those cards and letters flowing in.

• • • M E M 0 TO: FROM: SPI KWB DATE: 2 July 1977 RE: Basic plant sampling along the STX transect JCZ has raised animportant point regarding the planning for the sampling in STX and later on the other sites and cruises. Clearly, we can not issue the correct answers to the problem on the first try. We will need a few runs to work out the problems. In that spirit, I offer the following suggestions, expecting them to be corrected as conditions and problems demand. 1. 2. a. Remember that most of the measurements from a core will be subsample measurements. If we are to relate these back to the whole core, and thereby be able to relate them to the . entire area, we must know what the subsample represents. This must be clearly recorded somewhere. The general sample processing procedure for each core which I would suggest is as follows: Separate the seagrasses into species. Each of the following steps then relates to what should be done with each species. b. Separate the short-shoots, roots, and rhizomes. c. Note any evidence of flowering, seedlings, seeds in the sediment, etc. -d. Count the number of short-shoots, record. e. Clean the sediment from the rhizomes, dry on papers in the sun. Later, it will be necessary to do a 24 hour oven drying and to weigh and record the dry weight. If sample is large, divide the rhizomes into some fraction of the whole (record this!!!) based on the wet weight and carry on with the drying as above .

• • • SPI, 2 July 1977 f. Clean the sediment from the roots, dry on papers in the sun. Follow the same procedure as given in (e) for the rhizomes. g. Divide the short-shoot portion into two equal subsets based on the wet weight. h. Process one short-shoot sample for dry weights as above. Note that no attempt will be made to remove the epiphytes from this subsample (a correction will be made based on information from the other subsample.) i. From the remaining subsample of short-shoots, choose 20 leaves which are relatively intact and bag these for leaf width and area measurements. It may be possible to Xerox the bag at this stage while the leaves are fresh and then discard the original leaves (for the present, keep the leaves, however). j. Take a sample of short-shoots and count the number of leaves per short-shoot. Record . k. Take a sample of leaves (record the number of leaves) and process these for the epiphyte biomass as follows: i. freeze the leaves with dry ice. ii. place the frozen leaves in a freeze dryer. iii. strip the epiphytes from the freeze-dried leaves by using a thin, fine spatula while holding the leaves with a pair of curved forcepts. iv. collect the epiphytes on glassine weighing paper (9x9 or 12x12 inch sheets are a good size). v. it may be a good idea to save the leaves, if possible, so that they may be processed for the leaf area . The above procedure of xeroxing may work. Obviously, correction for shrinkage wi 11 be necessary. 3. I have not included any measurement of the rhizome lengths in this procedure. It may have to be added . 4. A coding form will have to be devised.
